The work of preparing International Standards is normally carried out through ISO technical committees. This method is applicable to all types of vegetable lecithin. The purpose of the method is to enable the analysis of lecithin under several regulations. Lecithin (Codex International Numbering System for Food Additives (INS) No. 322) is a generally permitted additive and the determination of the TIM is part of many specifications. The purity requirement with regards to toluene insoluble content is based on the method described herein. Toluene is the replacement for benzene, which is used in older methods, but must not be used for reasons of toxicity. 2 Normative references The following referenced document is indispensable for the application of this document. For dated references, only the edition cited applies. For undated references, the latest edition of the referenced document (including any amendments) applies. ISO 4793:1980, Laboratory sintered (fritted) filters; Porosity grading, classification and designation 3 Definition For the purposes of this International Standard the following terms and definitions apply. 3.1 Toluene Insoluble Matter (TIM) Quantity of those substances that are insoluble in toluene under the conditions of this method. The Toluene Insoluble Matter is expressed in grams per 100 grams. 4 Principle The sample is dissolved in toluene and filtered through a glass filtering crucible of defined pore size (P40). The insoluble residue is dried at (103 ± 2) °C and weighed. NOTE Glass filtering crucibles with other pore sizes give different results and shall not be used. 5 Reagents WARNING Attention is drawn to the regulations which specify the handling of hazardous substances. NOTE According to ISO 4793 the porosity G3 (G2) is denominated as P40 (P100). The pore size of 16 µm to 40 µm is recommended by JECFA (www.fao.org/ag/agn/jecfa-additives/specs/monograph4/additive-250-m4.pdf) for the determination of TIM. IMPORTANT Cleaning procedure of the glass filtering crucibles: Fill the ultrasonic bath with an phosphate-free alkaline cleaning solution2 for laboratory glassware with a concentration of 10 %. Put the glass filtering crucibles into the ultrasonic bath for 30 min. Wash the glass filtering crucibles with water, and if necessary repeat the cleaning step. Clean the glass filtering crucibles in the laboratory cleaning machine. Use each glass filtering crucible for 10 analyses only, as then the pores are blocked and can not be cleaned to a sufficient standard. 6.2 Drying oven, temperature adjustable at (103 ± 2) °C. 6.3 Desiccator with silica gel. 6.4 Glass beaker, of 150 ml capacity, tall form. 6.5 Filtering bottle. 6.6 Vacuum pump (for the filtration). 6.7 Analytical balance. 6.8 Measuring cylinder, of 50 ml capacity. 6.9 Glass rods of different size. 7 Sampling A representative sample should have been sent to the laboratory. It should not have been damaged or changed during transport or storage. Sampling is not part of the method specified in this International Standard. A recommended sampling method is given in ISO 5555 1. 7.1 Preparation of the test sample: The sample is heated to max. 60 °C avoiding local overheating, and is homogenized by powerful stirring. Specific treatments of the test sample (filtration, melting etc) shall be mentioned in the analytical report. 1 DURAN® filtering crucible , porosity 2, volume 30 ml, diameter 36 mm, Cat.
